摘要
目的:观察粉防己碱作用于EC9706细胞后细胞增殖、细胞周期及sPLA2-Ⅱa蛋白表达的变化。方法:15μmol/L的粉防己碱作用于EC9706细胞1、3和5d后,MTT法检测细胞增殖情况,流式细胞术检测细胞周期的变化,免疫细胞化学SP法检测sPLA2-Ⅱa蛋白表达水平的变化。以未经粉防己碱处理的细胞作为对照组。结果:粉防己碱作用1、3和5d后,EC9706细胞的增殖抑制率逐渐增高(F=48.740,P<0.001),G0/G1期细胞比例逐渐增高(F=21.140,P<0.001),而S、G2/M期细胞比例逐渐减低(F=13.740,9.370,P<0.05),但sPLA2-Ⅱa蛋白的表达水平无明显变化(F=1.416,P=0.265)。结论:粉防己碱能够抑制EC9706细胞的增殖,但对sPLA2-Ⅱa蛋白的表达无影响。
Aim: To observe the effects of tetrandrine on cell proliferation, cell cycle and expression of sPLA2-II a protein in EC9706 cells and explore the possible mechanism. Methods: EC9706 cells were treated with 15 p.mol/L tetran- drine for 1,3 and 5 d. Cell proliferation was detected by MTT method, and cell cycle was detected by flow cytometry. The expression of sPLA2- Ⅱ a protein was detected by SP immunocytochemical method. The control group was not treated. Re- suits: After being treated by tetrandrine for 1,3 and 5 d, the proliferation of EC9706 cells was inhibited, and the inhibition rate increased with time( F = 48. 740, P 〈 0. 001 ). Flow cytometry revealed that compared with control group the G0/GI phase cells increased(F = 21. 140, P 〈 0. 001 )and the S, G2/M phase ceils decreased significantly after being treated by tetrandrine for 1,3 and 5 d respectively( F = 13. 740,9. 370 ,P 〈 0.05 ). The expression of sPLA2- Ⅱ a protein level in con- trol group and the three experimental groups had no statistical difference( F = 1. 416 ,P = 0. 265 ). Conclusion : The prolif- eration of EC9706 cells can be inhibited by tetrandrine,but the expression of sPLA2-Ⅱ a protein in EC9706 cells is not in- fluenced by tetrandrine.
