摘要
目的探讨原花青素(PC)对大鼠发生脑缺血再灌注损伤脑组织中白介素-6(IL-6)和内皮素-1(ET-1)的影响。方法 40只健康Wistar大鼠随机分为假手术组、模型组、PC高剂量组、PC低剂量组4组。采用改良线栓法制备大鼠局灶性脑缺血再灌注模型,其中PC高、低剂量组大鼠术前0.5h和缺血2h时给予PC溶液150mg/kg、75mg/kg,而假手术组和模型组大鼠分别给予等体积的生理盐水。再灌注结束后,采用ELISA法测定脑组织中IL-6、ET-1水平。结果 PC高、低剂量组IL-6、ET-1水平低于模型组(P<0.05)。PC高、低剂量组脑组织中IL-6、ET-1水平比较差异有统计学意义(P<0.05)。结论 PC能够明显改善脑缺血再灌注损伤,这可能与其减轻炎症反应有关。
Objective To observe the influence of procyanidine (PC) on interleukin -6 (IL-6) and endothelin-1 (ET-1) in brain tissue after cerebral ischemia-reperfusion in rats. Methods Forty Wistar rats were randomly divided into 4 groups (the sham operation group, model group, PC high dose group and PC low dose group). The modified thread block method was used to establish the focal cerebral ischemia-reperfusion in rats, 75 or 150 mg/kg PC solution was given at half hour before operation and 2 hours after ischemia in the PC high dose group and PC low dose group, while equal normal saline was given in the sham operation group and model group. The levels c^f IL-6 and ET-1 in brain tissues after cerebral ischemia-reperfusion in rats were detected by ELISA method. Results Compared with the model group, the levels of IL-6 and ET-1 in brain tissues after cerebral ischemia-reperfusion were lower in the PC high dose group and PC low dose group (P〈0.05). There were statistical differ- ences in the levels of IL-6 and ET-1 between the PC high dose group and PC low dose group (P〈0.05). Conclusion PC can improve brain tissues after cerebral ischemia-reperfusion through lightening the inflammatory reaction.
