摘要
目的 :建立中药玄参中哈巴俄苷和肉桂酸的高效液相含量测定方法 ,提出特征性有效成分哈巴俄苷的含量限度建议。方法 :Ultrasphere 5 μmODS分析柱 (2 5 0mm× 4 6mm) ,流动相为乙腈和 1%醋酸水溶液 ,线性梯度洗脱 (2 0∶80→ 5 0∶5 0 ) (2 0min) ,流速为 1mL·min 1,室温 ,检测波长为 178nm。结果 :哈巴俄苷与肉桂酸回收率分别为 97 13%和 99 38% ,RSD分别为 0 80 %和 0 5 1% ,全国 2 0份商品饮片和 8份不同产地完整药材中哈巴俄苷含量为 0 0 41%~ 0 0 6 8%。结论 :本文方法快速、简便、准确 ,可用于玄参质量控制。建议玄参中哈巴俄苷含量应不低于 0 0 5 %。
Objectives:To establish an accurate method for the determination of harpagoside and cinnamic acid in Radix Scrophulariae(Xuan\|shen)and provide a proposition for the lowest content of the characteristic and active constituent (harpagoside).Methods:Gradient HPLC method was setup.The experimental conditions were as follows:Ultrasphere 5 μm ODS column(250 mm×4 6 mm),mobile phase:acetonitrile-water(ontaining 1 0% acetic acid)(20∶80→50∶50)(20 min),flow-rate 1 mL·min -1 ,room temperature,detection wavelength 278 nm.Results:The recoveries and RSD were 97 13%,0 8% for harpagoside and 99 38%,0 51% for cinnamic acid.28 samples of Xuan-shen(Radix Scro-phulariae)from different districts of China were quantitatively analysed and the contents of harpagoside and cinnamic acid in Xuan-shen were 0 041~0 244% and 0 012~0 068% respectively.Conclusion:The method was simple and accurate,and can be used for the quality control of Radix Scrophulariae.We propose that the content of harpagoside in the roots of Radix Scrophalariae must be no less than 0 05%.
