摘要
利用随机扩增多态性DNA(RAPD)技术 ,在DNA分子水平上对疣蝗 5个种群间的遗传差异进行了研究 .在 2 3个引物中有 6个引物出现稳定的扩增带 ,扩增总带数为 6 6条 ,其中 5 3条具多态性 ,占 80 3% ,平均每个引物 8 9条 .利用Nei&Li相似系数和UPGMA聚类法对结果进行分析 ,发现用该法检测到的遗传差异从南向北是连续性梯度差异 。
The genetic differences of 5 populations of Trilophidia annulata (Thunberg) are compared at DNA level using RAPD method. Of 23 10 base arbitrary primers that are tested, 6 are informative and yielded 66 bands, 53 (80.3%) of which are polymorphs bands with a mean 8.9 bands per primer. Nei′s similarity index and UPGMA are used for analyzing the genetic relationships among 5 populations. The results show that RAPD patterns are species specific and appear continuous variation from south to north populations T. annulata should be a monotypic species in the studied regions.
