摘要
用硫酸铵分级后再用phenyl-Sepharose CL-4B,DEAE-Sephadex A-50柱层析得到均一的人红细胞谷胱甘肽过氧化物酶(GSH-Px)。该酶在SDS聚丙烯酰胺凝胶电泳呈单一区带。用酶联免疫吸附法测定了克山病病区儿童和非病区儿童血GSH-Px活力;同时测定了大骨节病区和非病区儿童血GSH-Px活力。实验结果表明病区明显低于非病区儿童。
Glutathione peroxidase (GSH-px) was obtained and purified to hommogeneity by ammonium sulfate fractionation of human erythrocyte proteins followed by phenyl-sepharose CL-4B and DEAE-sephadex A-50 column chromatographies. The enzyme appeared a single band in SDS-polyacrylamide gel electrophoresis. Applied study showed that blood GSH-px activities in children from endemic areas of Keshan and Kaschin-Beck diseases were significantly lower than those from non-endemic areas.
