摘要
分别采用不同剂量的紫外线(UV)和甲基磺酸乙酯(EMS)诱变2种真眼点藻(波氏真眼点藻(Eustigmatos polyphem)和魏氏真眼点藻(Eustigmatos vischeri)),以氯霉素、红霉素、硫酸新霉素、硫酸卡那霉素为抗性筛选剂,利用抗生素最小致死浓度法筛选出抗性突变株。使用改良的BG-11培养基对反复筛选所获得的抗性突变株进行固、液培养,并以生物量干重、干藻粉的总脂含量、单位体积总脂含量、单位体积总脂产率为评价指标对抗性突变株的优劣进行评价。结果获得2株具稳定遗传特性、生长速度快、单位体积总脂产率高和具生产潜力的优良突变株。
Different doses of ultraviolet ray (UV) and ethyl methyl sulphonate (EMS) were used as mu- tagen to treat two eustigmatophyte microalgaes (Eustigmatos polyphem, Eustigmatos vischeri) respectively. According to the minimum antibiotics lethal concentration approach, chloromycetin, erythromycin, neomy- cin sulfate and kanamycin sulfate were chosen as resistance agent to screen mutant strains. The different mutant strains were cultured in modified solid and liquid BG - 11 mediums for assessing their growth and lipid accumulation. Dry biomass weight, total lipid content, volumetric total lipid content and volumetric total lipid productivity were employed to evaluate the properties of the selecting mutant strains. Finally, two excellent mutant strains with fast growth, high volumetric total lipid productivity, stable genetic char- acteristics and potentially commercial development value were obtained.
