An Aptamer-based Assay for the Detection of Lysozyme with Gold Nanoparticles by Surface Plasmon Resonance

摘要 首先建立了一种基于表面等离子体共振技术的适配体传感器检测溶菌酶。通过自组装的方法将1,4-苯二硫醇连接于芯片表面,将组装好的芯片用金纳米修饰,最后将溶菌酶适配体固定到修饰好的芯片上。结果表明,在0.2~20μmol/L范围内具有良好的线性关系,线性方程为Y=41.81+3.60 X,检出限为0.09μmol/L。对人血清样品检测,平均回收率为92.9%~99.6%。 In this work,an aptamer-based assay for the detection of lysozyme by surface plasmon resonance was described.Gold nanoparticles were anchored onto a sensor chip surface which was previously modified with self-assembled monolayers of 1,4benzenedithiol.Lysozyme aptamer was immobilized to the modified gold chip.The results showed that the linear range was from 0.2μmol / L to 20μmol / L,the linear equation was Y = 41.81 + 3.60X,the detection limit was 0.09μmol / L.The proposed method was applied to the determination of lysozyme in human serum samples,the average recoveries were 92.9%-99.6%.
