护卵汤对GnRHa超排卵大鼠卵巢FSHR和LHR蛋白表达的影响 被引量:4

Effects of Huluan decoction on the expression of FSHR and LHR in rats' ovary under GnRHa controlled ovarian hyperstimulation

摘要 目的观察护卵汤对超排卵大鼠卵巢促性腺激素受体———卵泡刺激素受体(FSHR)和黄体生成素受体(LHR)蛋白表达的影响,探讨其相关的作用机制。方法模仿人类超排卵长方案,对大鼠使用促性腺激素释放激素(GnRHa)降调节及尿促性腺激素(HMG)等药物超排卵(模型组),同时给予部分大鼠不同剂量的中药护卵汤(护卵汤低、中、高剂量组),以及未使用药物的自然周期大鼠为正常对照(正常组)。在注射绒毛膜促性腺激素(HCG)当天摘取卵巢组织,用免疫组化法检测FSHR和LHR的蛋白表达。结果 1、与正常组比较,模型组卵巢FSHR蛋白表达减少,差异有统计学意义(P<0.05);两组大鼠卵巢LHR蛋白表达的差异无统计学意义(P>0.05)。2、与模型组比较,护卵汤高剂量组卵巢FSHR和LHR蛋白表达均增加,差异有统计学意义(P<0.05);3、与模型组比较,护卵汤中剂量组卵巢LHR蛋白表达显著增加,差异有统计学意义(P<0.01),两组间卵巢FSHR蛋白表达差异无统计学意义(P>0.05);4、护卵汤低剂量组与模型组比较,卵巢FSHR和LHR蛋白表达差异无统计学意义(P>0.05)。结论护卵汤能改善GnRHa超排卵大鼠卵巢FSHR和LHR的蛋白表达,从而改善卵泡发育及卵巢反应性。 Objective To observe the effects of Huluan decoction on the expression of FSHR and LHR in rats' ovary under GnRHa controlled ovarian hyperstimulation(COH),and to discuss the mechanism of Huluan decoction in assisted reproductive technology(ART).Methods With the medications of GnRHa and HMG/HCG which imitates the human long period of COH,a number of rats were modeled and then treated with different doses of Huluan decoction(high dose,medium dose and low dose) and a number of rats without treatment were selected as normal group.The expression of FSHR and LHR in ovary were observed by immunohistochemistry on the day of HCG injection.Results 1.Compared with the normal group,the expression of FSHR in ovary in the model group was decreased significantly(P〈0.05) while there was no significant difference in LHR expression between the two groups(P〈0.05).2.Compared with the model group,the expressions of FSHR and LHR in ovary were significantly increased in the high dose group(P〈0.05).3.Compared with the model group,the LHR expression in ovary was increased significantly in the medium dose group(P〈0.01) while there was no difference in FSHR expression between the two groups(P〈0.05).4.There were no differences in the expression of FSHR and LHR in ovary between the model group and the low dose group(P〈0.05).Conclusion Huluan decoction can increase the expression of FSHR and LHR in ovary,which may improve the ovarian response and the follicular development in rats under GnRHa superovulation.
