摘要
目的:观察乌司他丁对哮喘小鼠BALF中细胞因子及嗜酸性粒细胞(eosinophil,EOS)的影响。方法:雌性SPF级BALB/c小鼠40只随机分为4组,即对照组、哮喘模型组、乌司他丁治疗组、地塞米松组,每组10只,除对照组外以卵蛋白致敏并吸入激发制备小鼠哮喘模型,治疗组均在每次哮喘激发前30min腹腔注射给药,激发并给药28d后处死小鼠。检测BALF中IL-2、IL-4、IL-5、IL-10、IFN-γ水平并计数EOS。结果:与哮喘模型组比较,乌司他丁治疗组小鼠BALF中EOS明显降低;IL-2、IL-10、IFN-γ明显升高(P<0.01),IL-4、IL-5明显降低(P<0.01)。结论:乌司他丁可能通过调节各种细胞因子的分泌及EOS的浸润,抑制哮喘小鼠气道炎症。
Objective To explore the effects of ulinastatin on cytokines and eosinophils in BALF from bronchial asthmatic mice.Methods 40 female BALB /c mice were randomly divided into four groups including control group,asthma model group,ulinastatin tgroup,and dexamethasone group,10 for each group.The rat model of bronchial asthma was established by intraperitioneal sensitization and stimulated with nebulized ovalbumin(OVA).The mice in each treatment group were administrated with ulinastatin or dexamethasone at 30 mins before every stimulation daily by peritoneal injections from the first asthma sensitization to the day before killing the mice,while the mice in the control group were administrated with physiological saline.The mice were killed 28 day after treatment.IL-2,IL-4,IL-5,IL-10,and IFN-γ in BALF were detected,and the number of EOS was counted.Results As compared with the asthma model group,the number of EOS significantly reduced(P〈0.01),levels of IL-2,IL-10,and IFN-γ significantly increased,and levels of IL-4 and IL-5 significantly declined in ulinastatin group(P〈0.01).Conclusions Ulinastatin possibly suppresses airway inflammation in bronchial asthmatic mice by regulating secretion of cytokines and infiltration of eosinophils.
