摘要
目的探讨丰富环境对局灶性脑梗死大鼠运动功能、学习记忆及Nogo-A蛋白表达的影响。方法手术组予线栓法制作SD大鼠右侧大脑中动脉缺血模型,术后24 h随机分为丰富环境组和标准环境组,分别饲养于普通鼠笼及丰富环境鼠笼;另设假手术组,不线栓大脑中动脉,其余步骤与手术组相同。各组分别在术后7、14、28 d分别用旷场实验、水迷宫实验进行行为学分析。术后28 d行为学分析后取脑,行免疫组化观察各组Nogo-A阳性细胞表达情况。结果术后28 d丰富环境组的运动总路程[(4 360±854)cm]与标准环境组[(2 362±694)cm]比较,差异有统计学意义(P<0.001)。定位航行实验,丰富环境组在术后14、28 d的潜伏期[分别为(33.0±5.8)s、(24.5±3.1)s]与标准环境组[分别为(46.6±6.0)s、(35.6±4.7)s]比较,差异有统计学意义(P<0.001)。空间探索实验,丰富环境组在术后28 d的穿越平台次数[(6.6±1.9)次]与标准环境组[(3.2±1.5)次]比较,差异有统计学意义(P<0.001)。丰富环境组梗死灶周围Nogo-A阳性细胞数(86.7±6.7)与标准环境组(117.5±8.3)比较,差异有统计学意义(P<0.001)。结论丰富环境能促进脑梗死大鼠的运动能力、记忆力的恢复。丰富环境能降低脑梗死大鼠梗死灶周围Nogo-A蛋白表达,从而促进神经再生。
Objective To investigate the effects of rich environment on the athletic function, learning and memory functions as well as the expression of Nogo - A protein in rats with focal cerebral infarction. Methods Right middle cerebral artery occlusion (MCAO) model of SD rats was constructed, which were randomly divided into enriched environ- ment group and standard environment group 24 h after operation. A sham group was also set up. Open field test and Morris water maze test were performed on the 7th, 14th and 28th day. The rats were sacrificed on the 28th day for assessment of Nogo - A in brain. Results On the 28th day, the total distance of enriched environment group [ (4 360 ± 854 ) cm ] was significantly longer than standard environment group [ (2 361 ±694) cm] (P 〈0. 001 ). On the 14th and 28th day, the latency of enriched environment group [ ( 33.0 ±5.8 ) s and ( 24. 5 ±3.1 ) s ] were significantly shorter than those in standard environment group [ (46. 6 ±6. 0 ) s and ( 35.6 ±4. 7 ) s, P 〈 0. 001 ]. On the 28th day, the across platform frequency of spatial probe test of enriched environment group ( 6. 6 ±1.9) was significantly higher than standard environment group ( 3.2 ±1.5 ). Around the infarction region, significant increased Nogo - A positive cell count was observed in enriched environment group. Conclusion Rich environment can promote the recovery of athletic ability and memory function in rats with focal cerebral infarction, probably via down - regulating the Nogo - A protein expression around the infarction, which promotes nerve regeneration.
