摘要
目的建立茶枝柑皮提取物中黄酮类成分的高效液相色谱含量测定方法。方法采用PhenomenexLuna C_(18)(250 mm×4.6 mm,5μm)色谱柱,检测波长为283 nm和330 nm,以乙腈-0.2%(体积分数)乙酸水溶液为流动相梯度洗脱,柱温为25℃。结果橙皮苷、川陈皮素和橘皮素分别在8.240~164.8μg/mL(r=0.999 7)、8.030~160.6μg/mL(r=0.999 8)和8.210~164.2μg/mL(r=0.999 8)范围内呈良好线性关系。回收率分别为97.3%、99.1%和97.6%,RSD分别为2.6%、0.9%和1.3%。结论本方法简便快捷、重复性好、准确度高,为茶枝柑皮提取物质量标准的建立提供了有力依据。
Objective To establish a HPLC method to determine flavonoids from Citrus reticulata ' Chachi' peel extract. Methods Hesperidin, nobiletin and tangeretin were determined on Phenomenex Luna C18 (250 mm× 4. 6 mm, 5 μm) column and detected at 283 nm and 330 nm. The mobile phase was consisted of acetonitrile and 0.2% acetic acid. The column temperature was 25 ℃. Results The linear range of hesperidin was within 8. 240 - 164.8 p,g/mL ( r =0.999 7), and the recovery was 97.3% with RSD = 2.6%. That of nobiletin was within 8. 030 - 160.6 p,g/mL (r =0.999 8), and the reeovelT was 99. 1% with RSD =0.9%. That of tangeretin was within 8. 210 - 164. 2 μg/mL ( r =0. 999 8), and the recovery was 97.6% with RSD = 1.3%. Conclusion This method is simple and accurate, which may provide a reference for the quality control of Citrus reticulate ' Chachi' peel extract.
