摘要
β-酮脂酰-ACP合酶——KAS是脂肪酸合成中的一种缩合酶,主要是在缩合反应中起作用。研究以近成熟的油桐种子为材料,根据GenBank中蓖麻等已知的β-酮脂酰-ACP合酶基因KASⅢ的序列设计简并引物,利用PCR、RACE等技术克隆得到了油桐KASⅢ基因的全长cDNA序列。该基因序列全长1 901 bp,开放阅读框为1 209 bp,编码403个氨基酸,通过与其他物种的KASⅢ氨基酸序列比对,油桐KASⅢ基因与麻疯树的同源性最高,达90%。软件预测表明,KASⅢ蛋白等电点为5.78,存在跨膜结构域,整个蛋白质基本上是表现出亲水性的。将该基因命名为vf-kasⅢ。
Beta-ketoacyl-ACP synthase is a fatty acid synthesis under the condensation effect of enzyme, plays a role in the condensation reaction. The primer was designed according to the KAS Ⅲ sequences recorded in genbank. Full-Length cDNA of KAS Ⅲ was obtained by cloning it with PCR, RACE, etc. methods. It was 1 901 bp in length and contained a 1 209 bp ORF (open reading frame) encoding a peptide of 403 amino acids, amino acid sequence highest similarity(90% ) to brane spanning domain was alignment with other species showed that the KAS Ill of Verniciafordii displayed the Jatropha curcas.
