胰岛素生长因子-1在糖尿病大鼠膀胱和腰骶背根神经节中的表达 被引量:1

Expression of insulin-like growth factor-1 in the bladder and lumbosacral dorsal root ganglia of diabetic rats

摘要 目的 观察胰岛素生长因子-1(IGF-1)在糖尿病性膀胱病的发生发展中的作用.方法 将35只SD大鼠随机分为两组,对照组为15只,糖尿病组20只;建立糖尿病大鼠模型,11周后分别取两组大鼠膀胱及腰骶背根神经节组织,苏木素-伊红(HE)染色,光镜下观察.常规免疫组织化学方法检测IGF-1在大鼠膀胱及腰骶背根神经节组织的表达.结果 对照组血糖明显低于糖尿病组(4.7333 ±0.6172比29.5200±1.6224,P<0.05),光镜下见大鼠膀胱逼尿肌结构排列紊乱,肌间隙明显增宽,肌束间血管充血,逼尿肌细胞肥大,形态不规则.免疫组织化学显示,糖尿病组中IGF-1在膀胱及腰骶背根神经节中的表达明显下降(P<0.01).结论 IGF-1在糖尿病大鼠的膀胱及腰骶背根神经节中表达均明显下调,在糖尿病性膀胱病的病变发生及发展中起着重要作用. Objective To explore the function and mechanism of insulin-like growth factor-1 (IGF-1) in the diabetic bladder.Methods Thirty-five male Sprague-Dawley (SD) rats were divided into two groups randomly,including 15 in control group,and 20 in diabetic group.Rat diabetic models were made through injection of STZ.After 11 weeks,the histologic changes of the bladder and lumbosacral dorsal root ganglia (DRG) in rats were observed under light microscopy by hematoxylin and eosin (HE) staining.The expression of IGF-1 was detected by using immunohistochemistry.Results The blood sugar level in diabetic group was significantly higher than that in control group (29.5200 ± 1.6224 vs.4.7333 ±0.6172,P < 0.01).Immunohistochemical resuhs revealed that the expression of IGF-1 in detrusor cells and DRG of diabetic group was significantly decreasedas compared with control group (P < 0.01) ; DRG:(P < 0.01).Conclusion IGF-1 expression in the bladder tissue and DRG of diabetic rats was significantly reduced,which may play an important role in the pathogenesis of diabetic cytostopathy.
