摘要
由于没有明确的形态差异 ,香菇的 3种不亲和交配反应即A =B =,A =B≠和A≠B =难以互相区别。本研究将OWE SOJ技术及核迁移试验用于香菇的交配型分析。结果表明 ,应用OWE SOJ技术 ,可以观察到 3种不同的菌落形态 :由A≠B≠配对形成的具锁状联合的绒毛状菌落 ,由A≠B =配对形成的无锁状联合的栅栏型菌落及由A =B =或A =B≠配对形成的无锁状联合的绒毛状菌落。通过一个附加的常规交配试验 ,A =B =和A=B≠反应易于互相区别开来。核迁移试验表明 ,核迁移出现于A =B≠反应中而不出现于A≠B =反应中。这一结果与OWE SOJ试验的结果是一致的。因此 ,在常规交配型分析的基础上 ,应用OWE SOJ技术及核迁移试验以准确地鉴定样本中所有单核体的交配型因子是可行的。
The three incompatible mating interactions, A=B=, A=B≠and A=B=, are hardly distinguishable from each other because of the absence of definite morphological differences in Lentinula deodes .In this study, OWE?SOJ technique and nuclear migration test were applied to the analysis of mating type in L.edodes .The result indicaed that by using OWE?SOJ technique, three different colony morphologies could be observed: a fluffy colony with clamp connection formed by the pairing of A≠B≠, a barrage colony without clamp connection formed by that of A≠B= and a fluffy colony without clamp connection by A=B= or A=B≠. A=B=and A=B≠interactions can be distinguished easily from each other by an additional ordinary mating test. The nuclear migration test showed that nuclear migration occurred in A=B≠ interaction but did not occur in A≠B= interaction. This result was consistent with that of the OWE?SOJ test. Therefore it is feasible to apply OWE?SOJ technique and nuclear migration test to determine mating type factors of all monokaryons in a sample accurately on the basis of routine mating type analysis.
