猴头菌液态发酵米糠工艺的优化 被引量:5

Optimization of liquid culture of Hericium erinaceum in medium with extract of rice bran

摘要 为高效转化米糠生产食用菌粗多糖,拟采用猴头菌发酵米糠得到猴头菌丝原料。以米糠为基础培养基进行猴头菌液体发酵,在对培养基成分和发酵条件进行单因素实验的基础上,采用响应面法对米糠浓度、蛋白胨添加浓度、装样量三个主要因素进行优化。优化实验的其他控制参数为:葡萄糖20g/L、培养基pH5.0、KH2PO41.5g/L、MgSO40.75g/L,接种量10%、培养温度28℃、培养时间7d,得到优化条件为:250mL三角烧瓶装样150mL,蛋白胨浓度9.8g/L,米糠浓度23.3g/L,实验结果猴头菌菌丝干重为(11.28±0.082)g/L。 To make efficient use of the rice bran and to get mushroom mycelium polysaccharides,Hericium erinaceum was used to ferment rice bran to product polysaccharides. Hericium erinaceum was fermented in liquid medium with extract of rice bran and the medium components and culture conditions were investigated through single-factor experiment. By Box-Behnken experiments ,in which Glucose 20g/L, KH2PO4 1.5g/L, MgSO4 0.75g/L,inoculation volume 10% ,pH5.0,culture time 7d,temperature 28℃ were fixed,concentration of rice bran,peptone conoentration and loading volume were optimized. The optimal value were 23.3g/L,9.8g/L and 150mL(250mL flask) respectively. The optimal dry weight of mycelium was 11.28g/L.
