摘要
为保护资源和实现栽培,以根茎段为材料,采用试管苗培养的方法,对汉城细辛进行了试管苗培养、试管苗繁殖培养、微型根茎扦插及试管苗移栽和定植的研究.结果证明:1/2MS+蔗糖12 g/L+ABT2号1.2 mg/L+IBA0.4 mg/L是试管苗培养和试管苗根茎培养的适宜培养基;1/2MS+蔗糖10 g/L+ABT2号1.2 mg/L+IBA0.5 mg/L+IAA0.1 mg/L是试管苗根茎的繁殖培养适宜培养基.微型根茎段成活率为97.0%;试管苗移栽成活率为98.25%;定植成活率为97.8%.定植后保持了野生汉城细辛的植物学性状.
In order to preserve the wild resources and realize cultivation,with the method of tissue culture, the rootstocks of Asarum sieboidii var seoulense Nakai were used as materials to research. The study consists of the cultivation of tube seedlings, propagation of tube seedlings, cuttings for miniature rhizome segments,transplanting and colonization of tube seedlings. The results demonstrated that 1/2MS+ sucrose 12 g/L+ABT 11 1.2 mg/L+ IBA 0.4 mg/L was the ideal medium for cultivation of tube seedlings, and 1/2MS + sucrose 10 g/L + ABT II 1.2 mg/L + IBA0.5 mg/L + IAA0.1 mg/L was the ideal medium for propagation of tube seedlings. The survival rate of miniature rhizome segments was 97.0%. The survival rate of transplanted tube seedlings was 98.25% and colonization survival rate was 97.8%. The colonization of plantlets maintained all biological traits of wild plant.
