摘要
目的探讨四君子汤灌胃对慢性左心衰模型大鼠心功能的影响,以心衰大鼠为切入点探讨"心脾相关"。方法 SD雄性大鼠腹主动脉缩窄法8周构建慢性左心衰模型,血流动力学指标确定建模成功。成模大鼠随机分为四君子汤高、低剂量组、模型组、卡托普利组。另设空白对照组。药物干预组常规喂养,并按剂量[高剂量组相当于11.88g生药/(kg·d)、低剂量组相当于2.97 g生药/(kg·d)]灌胃给药,每日1次,连续1月,再次测量血流动力学指标,取出心脏,分离并称取左右心室重量,心肌组织常规HE染色,光镜下观察心肌细胞及间质变化。结果模型组心功能较其他各组降低,心肌肥厚指数增加(与正常组比较P<0.05),血流动力学指标与正常组比较(P<0.05)有显著差异,四君子汤高剂量组可明显改善心衰大鼠心功能,心肌肥厚指数与模型组比较(P<0.05),血流动力学指标与模型组比较有显著差异(P<0.05)。光镜下可见四君子汤高剂量组较模型组心肌纤维排列紊乱、肥大状况改善,间质水肿,充血状况改善。结论四君子汤可以改善慢性左心衰模型大鼠的心功能,初步验证了"心脾相关"。
Objective To investigate the effect of Sijunzi Decoction on cardiac function of rats after chronic heart failure and its mechanism. Methods To establish rat model of heart failure by partially banding abdominal aorta for 8 weeks. The heart failure rats were randomly divided into 4 groups : Sijunzi Decoction preconditioning group ( High and low doses ) , model group, positive control group ,furthermore, 1 Sham operation group was established, After 1 month, myocardial hypertrophy index, cardiac function were determined, and the myocardial cells and interstitial change were observed with Microscope after myocardial tissue HE stai- ning. Results In Sijunzi Decoction preconditioning group, the cardiac function was markedly improved, and the myocardial hyper- trophy index was reduced markedly compared with the model group. Conclusion The cardiac function and ventricular remodeling are improved profoundly in heart failure rats treated with Sijunzi Decoction.
