摘要
目的 :建立测定人血浆中水飞蓟宾浓度的反相高效液相色谱法。方法 :血样在pH 8 5条件下经乙醚提取后 ,采用 μBondapakC18(30 0mm× 4 6mm)色谱柱 ,甲醇 -乙腈 -磷酸盐缓冲液 (pH 3 0 ) (2 9∶2 0∶5 1)为流动相 ,以萘酚为内标 ,检测波长 2 88nm。结果 :水飞蓟宾在 3 6 95~ 2 36 5ng·mL-1范围内线性相关 (r =0 9998) ,方法回收率为 96 5 % ,日内、日间RSD均小于 2 3% (n =3)。结论 :方法简便快速、灵敏度高 。
Objective:To establish a RP-HPLC method for the determination of silybin in human plasma.Method:The silybin was extracted from plasma at pH=8\^5 by ether.The mobile phase consisted of methanol,acetonitrile and phosphate buffer (pH 3\^0),detected in 288 nm.Results:The calibration curve was linear in the range from 3\^695 to 236\^5 ng·mL -1 with r=0\^999 8. The recovery was 96\^5%.\%RSD\% within a day and between days were less than 2\^3%.Conclusion:This method was simple,specific and accurate.The utility of the method for human pharmacokinetic studies was demonstrated.
