摘要
[目的]探索骨关节炎(osteoarthritis,OA)模型的病理改变。[方法]切断SD大鼠右侧膝关节前交叉韧带(ACLT),自然喂养,于术后3、7、14、28、56 d处死大鼠,取左右全膝,观察全膝形态改变、膝关节X线影像学改变、关节滑膜ED1和软骨结构改变。[结果]术后3、7 d滑膜单克隆抗体ED1阳性渗出性细胞明显增多;第7 d出现早期软骨损害:软骨基质减少,表层带软骨细胞死亡数量减少及软骨细胞肿胀,表层带纤维化等改变;28d后肉眼观察可见全膝形态明显改变,膝关节骨密度增高及滑膜纤维化增生等后期病理变化。[结论]SD大鼠前交叉韧带切断能反映骨关节炎(OA)的病理变化,是较理想的骨关节炎(OA)动物模型。
[ Objective] To find out the pathological changes in osteoarthritis models. [ Method] Transection of anterior cruciate ligament on the right knee joint (ACLT) was performed on SD rats. The SD rats were naturally fed and then killed. Both the total left and right knee joints were taken out on the 3 rd,7,14,28 th and 56 th day after the operations. The changes of the knee morphology,X- ray image, synovial ED1 and joint cartilage structure were observed on both the left and the right knee joints. [ Result] The number of monoelonal antibody (ED1) positive cells in synovium was found out increased significantly on the 3 rd and the 7 th day after operatioin. Early cartilage damage with reduced cartilage matrix, swollen and dead cartilage cells and superficial cartilage fibrosis were detected on the 7 th day, The total knee morphological changes were observed with increased bone density and synovial hyperplasia on the 28th day. [ Conclusion] The pathological changes of osteoarthritis can be simulated after anterior erueiate ligament was transected in SD rats and it is recommended an ideal animal model for osteoarthritis study.
