摘要
目的考察贯叶金丝桃提取物对人鼻咽癌的体外、体内光动力效应。方法对体外培养的CNE-2细胞分别加入不同纯度贯叶金丝桃提取物(高、中、低纯度)及纯品,光照处理后,MTT法测定细胞生长抑制率;选取抑制效果最好的提取物,以血卟啉为阳性对照,观察其对移植性鼻咽癌的光动力效应。结果高纯度金丝桃素提取物对CNE-2细胞的增殖抑制率最高,其半数抑制质量浓度为(0.093±0.004)μg/mL。体内药效学研究,高纯度金丝桃素提取物高剂量组(5 mg/kg)、低剂量组(2.5 mg/kg)的相对肿瘤增殖率分别为38.2%、59.7%,血卟啉组(5 mg/kg)的相对肿瘤增殖率为56.3%,均显著小于阴性对照组(P<0.01)。结论高纯度金丝桃素提取物能有效抑制体外、体内鼻咽癌细胞的增殖,具有良好的开发应用前景。
AIM To study the photodynamic effect of hyperium perforatum extract on human nasopharyngeal carcinoma in vivo and in vitro. METHODS Human nasopharyngeal carcinoma cells CNE-2 cultured in vitro were exposed to hypericin and different purity of extracts, then irradiated by LED light. MTT assay was used to detect the growth inhibition rate. Photodynamic effect on CNE-2 was observed compared to hematoporphyrin as positive control. RESULTS High parity hypericin extract had the best inhibitory effect in vitro, The medium effective concentration was (0.093±0.004)μg/mL. The relative rate of tumor reproduction (T/C % ) in high dose group and low dose group were 38.2% and 59.7% respectively , and in the positive control group was 56. 3%. The re- suits showed strong statistic differences (P 〈 0. 01 ) when compared with the blank group. CONCLUSION High purity hypericin extract has obvious photodynamic effect on human nasopharyngeal carcinoma in vitro and in vivo.
