消瘰颗粒醇提干浸膏中黄芩苷含量的HPLC法测定 被引量:3

摘要 采用高效液相色谱法,以AgilentEclipseXDB—C18柱(4.6mm×15cm,5μm)为色谱柱,甲醇-水-磷酸(55;45:0.2)为流动相,流速为1.0mL·min-1,检测波长为280nm,进样量为5~10μL,柱温为30℃.在给定的色谱条件下,黄芩苷与相邻峰分离度大于1.5,阴性对照试验无干扰.黄芩苷进样量在0.192~1.283μg之间与峰面积呈良好线性关系,回归方程Y=-0.3245+53.295X,r=0.9998.平均加样回收率为101.3%,RSD=1.85%.该法操作简便,结果准确,重现性好,可用于消瘰颗粒剂的醇提取工艺研究及中间体质控. To establish a method for determination of baicalin in dried ethanol extract of Xiaoluo granule, a HPLC method was set up. The sample was separated and analyzed with the Agilent Eclipse XDB--C18 column (4.6 mm× 15 cm, 5μm) as chromatogram column, and the methanol-- water-- phosphic acid solution (55 : 45 : 0.2) as mobile phase. The flow rate was set at 1.0 mL min-1 , the detective wavelength at 280 nm, the injection volume was 5-10 μL, and the column temperature was 30 ℃. With this chromatographic condition, the baicalin peak in dried ethanol extract sample of Xiaoluo granule chromatogram could be separated from other ingredient peaks completely. The calibration curve was linear at a ranges of 0. 192-1. 283μg with r = 0. 9998, and the average recovery from sample was 101.3% and RSD = 1.85%. This method is simple, accurate, re- producible, and can be used for extract preparation and intermediate quality control of Xiaoluo granule.
