注射用炎琥宁对脂多糖发热模型家兔解热机制的研究 被引量:6

摘要 目的研究注射用炎琥宁对脂多糖发热模型家兔血清中细胞因子(TNF-α、IL-1β、IL-6、cAMP、PGE2)和下丘脑中升温介质(cAMP、PGE2)的影响。方法建立家兔脂多糖(10 EU/kg)发热模型,监测不同时间点注射用炎琥宁对脂多糖发热模型家兔体温影响,绘制平均升温曲线;模型升温峰值时收集指标,ELISA法检测血清中细胞因子、下丘脑中升温介质变化。结果注射用炎琥宁能显著降低脂多糖发热模型家兔的体温,且量效关系较显著;注射用炎琥宁能显著抑制发热模型家兔血清中IL-6、cAMP、PGE2增加和下丘脑中cAMP、PGE2的释放。结论注射用炎琥宁对脂多糖发热模型家兔有较好的解热作用,其解热效果可能与其抑制外周IL-6、cAMP、PGE2增加和下丘脑cAMP、PGE2释放有关。 Objective To explore the effects of Potassium Sodium Dehydroandroan Drographolide Succinate for injection (PSDDSI) on the cytokines (TNF-α, IL-1β, IL-6, cAMP, and PGE2) in serum and the fever neurotransmitters (cAMP, PGE2) in hypothalamus of rabbits in lipopolysacchride (LPS)-induced febris response. Methods he LPS-induced febris response in rabbits (10 EU/kg) was established. The body temperature of rabbits administered with PSDDSI in LPS-induced febris response was detected at different time points, and an average temperature rising curve was drawn. The indicators at warming peak time of the model were collected, and the contents of cytokines in the serum and fever neurotransmitters media in the hypothalams were measured by ELISA-Kits method. Results PSDDSI could significantly reduce the body temperature of rabbits in LPS-induced febris response, and the dose-effect relationship was very significant; PSDDSI could significantly inhibit the release of IL-6 and cAMP in serum and PGE2 and cAMP in hypothalamus of rabbits in LPS-induced febris response. Conclusion PSDDSI has a good antipyretic effect on LPS-induced febris response in rabbits. The mechanism may be related to the inhibition of cytokines in the serum and the fever neurotransmitters in the hypothalamus.
