摘要
目的:探讨CD151在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中的表达及意义。方法:免疫组化SP法检测云南省肿瘤医院2006-01-01-2011-12-31收治的78例NSCLC和22例良性肺病变组织中CD151蛋白的表达,并结合临床资料进行分析。RT-PCR法检测正常支气管上皮细胞株BEAS-2B、肺腺癌细胞株A549、云南宣威肺癌细胞株XWLC-05和云南个旧肺鳞癌细胞株YTMLC中CD151mRNA的相对表达量。结果:CD151在NSCLC组织中表达阳性率为57.7%(45/78),高于对照组的18.2%(4/22),χ2=10.72,P=0.001。CD151蛋白表达与性别、年龄、是否吸烟、T分期、TNM分期、肿瘤细胞分化程度以及PS评分无明显相关性,P值均>0.05;与病理分型(χ2=6.312,P=0.043)和淋巴结数(χ2=7.386,P=0.007)有关。生存分析提示,CD151蛋白阳性表达预后差;Cox回归分析表明,CD151蛋白是影响NSCLC预后的独立危险因素,P=0.014。荧光定量PCR结果显示,CD151mRNA在细胞株中的表达为BEAS-2B>A549>XWLC-05>YTMLC。结论:CD151蛋白可以作为判断NSCLC预后的参考之一。CD151mRNA在细胞株A549和XWLC-05中高表达,为进一步研究肺癌高发区特殊环境作用下发病机制以及预防、治疗提供新的思路。
OBJECTIVE:To investigate the clinical significance of CD151 expression in non-small cell lung cancer. METHODS : Immunohistochemical staining of SP was applied in the specimens of 78 NSCLC and 22 benign pulmonary dis- eases to investigate the expression of CD151 protein from January ls~,2006 to December 3lth,2011 in Yunnan Province Tumor Hospital. The correlation between the expression of CD151 and clinicopathologic features of NSCLC was analyzed. RT-PCR was applied to explore the expression of CD151 mRNA in BEAS-2B, A549, Xuanwei lung adenocarcinoma cell lines XWLC-05 and Gejiu lung squamous carcinoma cell lines YTMLC. RESULTS: The positive rate of CD151 protein in specimens of NSCLC was 57.70/oo ,which was higher than 18.2% of benign pulmonary diseases (X2 =10.72,P=0. 001). Overexpression of CDJ51 protein in NSCLC was not correlated with sex, age, smoking, T-stage,TNM-stage, differentia- tion and performance status, but was correlated with histology ( X2 = 6. 312, P = 0. 043) and N-stage ( X2 = 7. 386, P = 0. 007). Kaplan-Meier survival analysis and log-rank test suggested that overexpression of CD151 protein had poor prog- nosis. Cox regression suggested CD151 protein expression was independent prognostic factor of NSCLC(P^0. 014). Real- time PCR showed the expression of CD151 mRNA was BEAS-2B〉A549〉XWLC-05〉YTMLC. CONCLUSIONS: Over- expression of CD151 protein in NSCLC is correlated with poor prognosis and it can be served as a prognostic factor of NSCLC. There is overexpression of CD151 mRNA in A549 and XWLC-05 ,which could provide new ideas for the study of pathogenesis, prevention and treatment of Iung cancer developed in special environment.
