摘要
目的探讨三种主要人类白细胞抗原G(HLA-G)受体,即CD158d、CD85j和CD85d,在胶质母细胞瘤中CD4+T细胞的表达情况。方法分离胶质母细胞瘤和正常脑组织中CD4+T细胞利用佛波酯和离子霉素进一步激活CD4+T细胞。利用流式细胞术分析CD4+T细胞三种HLA-G受体的表达。结果胶质母细胞瘤中CD4+T细胞表面CD158d、CD85j和CD85d的表达阳性率较正常脑组织明显升高(P<0.05),应用佛波酯和离子霉素刺激后,这三种受体表达水平显著增高(P<0.05)。结论胶质母细胞瘤中CD4+T细胞存在HLA-G受体表达的上调现象,高表达HLA-G受体可能与肿瘤中T细胞功能抑制并最终引发肿瘤的免疫逃逸有关。
Objective To investigate the expressions of human leukocyte antigen (HLA)-G receptors including CD158d, CD85j and CD85d in cultured tumor-infiltrating CD4^+ T cells isolated from human glioblastomas. Methods The CD4^+ T cells which were isolated from 16 samples of human glioblastomas and 10 samples of normal brain tissues were cultured in DMEM culture. These cells were then stimulated with ionomycin (1 pg/ml) and PMA (10 ng/ml). The expression levels of CD158d, CD85j and CD85d were detected by flow 'cytometry. Results The purity of CD4~ T cells was more than 95%. Five hours after the stimulation with ionomycin and PMA, The positive rate of CD69 was more than 95%. The positive expression rates of CD158d (16.3%±4.2%), CD85j (11.5%+2.9%) and cd85d (31.9%± 12.4%) in CD4^+ T cells isolated from the glioblastomas samples were significantly higher than those (5.3%± 1.7%, 3.6%±0.7% and 12.7%±3.8%, respectively) from the normal brain tissues (P〈0.01). The positive expression rates of CD158d (15.3%±4.2%), CD85j (11.5%±2.9%) and cd85d (31.9%±12.4%) in CD4+ cells isolated from the glioblastomas samples before the stimulation with ionomycin and PMA were significantly lower than those (29.5%±6.8%, 19.3%±4.1% and 44.9%± 11.2%, respectively) after the stimulation (P〈 0.01). Condnsions HLA-G receptos including CD158d, CD85j and CD85d are up-regulated in tumor-infiltrating CD4^+ T cells isolated from human glioblastomas. It is suggested that the HLA-G receptors may be involved in the immunosupression and immunoescape of human glioblastomas.
