摘要
观察肾上腺髓质素前体N端20肽(PAMP)和肾上腺髓质素(ADM )对血管紧张素II(AngII)诱导的血管平滑肌细胞(VSMC)增殖的影响。用测定培养的大鼠VSMC3H-TdR掺入和蛋白激酶C(PKC)活性的方法。结果发现:10-8mol/L AngII刺激培养的VSMCs 3H-TdR掺入增加2.68倍(P<0.01)、PKC活性增加1.02倍(P<0.01)。而10-9mol/L-10-7mol/L的PAMP或ADM与10-8mol/L AngII同时孵育,则显著抑制AngII的上述作用(P<0.01)。且两者的抑制作用无明显差异。PAMP和ADM均有抑制AngII所致的VSMC增殖作用,可能是通过抑制信号传导系统中的PKC活性来实现的。PAMP和ADM 与AngII相互作用,共同调节VSMC的增殖,从而维持循环系统功能的稳态。
To observe the effect of proadrenomedullin N-term inal 20 peptide (PAM P)and adrenomedullin (ADM)on AngII-stimulated proliferation of vascular smooth m uscle cells(VSMCs). The 3H-TdR incorporation and intracellular PKC activity were measured in the cultured rat VSMCs. This study demonstrates that AngII (10-8 mol/L)significantly increased the VSMCs 3H-TdR inco rporation by 2.68-fold( P <0.01) and PKC activity by 1.02-fold( P <0.0 1).PAMP and ADM ( 10-9~10-7 mol/L) markedly inhibited the AngII -s timulated 3H-TdR incorporation and PKC activation in a c oncentration-dependent manner .Conclusion: PAMP and ADM possess the ability to inhibit the AngII-induced proliferation of VSMCs through inhibiting the PKC act ivity. It suggests that PAMP,ADM and AngII may play a role to maintain the circ ulatory homeostasis by interact of each other.
