摘要
对诱变获得展青霉LD100-1菌株进行固态培养工艺优化,为提高灰黄霉素产量提供基础数据。结果表明,LD100-1最佳固态培养参数为以晚籼米、NaNO3作为碳源和氮源,初始料水比=1∶0.75,添加1.1%NaCl作为Cl-来源,再添加适量疏松剂。以培养44h的种子液接种,接种量为10%。培养过程采用变温发酵1d^7d、28℃,8d^14d、25℃。工艺优化后灰黄霉素效价达160000μg/g,提高了60%。
Solid cultivation process optimization of Penicillium patulum LD 100-1 which has been induced mutation to supply basic data for approving griseofulvin production.The results showed that the optimum conditions were as follows:indica rice and NaNO3 as carbon and nitrogen sources,the initial ratio of material to water 10.75,addition of 1.1% NaCl as Cl-sources with bulking agent.Vaccinating 10% of the seed liquids which have been fostered for 44h; using variable temperature fermentation of 1d~7d 28℃ and 8d~14 d 25℃.The griseofulvin titer has increased by 60000μg/g(60%) after optimization.
