摘要
以抗脱氧雪腐镰刀菌烯醇(Deoxynivalenol,DON)单克隆抗体为基础,建立了胶体金免疫层析试纸条检测方法,用于小麦和玉米中DON的快速检测。采用柠檬酸三钠还原法制备胶体金,标记抗DON单抗并喷于玻璃纤维垫上,DON-BSA和驴抗鼠二抗均匀划于NC膜上检测线和质控线位置,依次将样本垫、结合垫、NC膜和吸水纸组装切割成试纸条并装入检测卡中。测试结果表明:该胶体金试纸条对纯标准品和人工污染样品的检测灵敏度分别为100μg/kg和500μg/kg,与除15-乙酰脱氧雪腐镰刀菌烯醇外的21种生物毒素均无交叉反应,重复性和稳定性均良好,对实际样品的检测结果与液相色谱串联三重四级杆质谱(LC-MS/MS)比对结果相符。该检测方法非常适合用于现场的快速筛查,具有良好的应用前景。
Based on anti-deoxynivalenol(anti-DON) monoclonal antibody, colloidal gold immuno- chromatography strip test was developed for the rapid detection of deoxynivalen in wheat and maize. Anti-DON monoclonal antibody coupled with colloidal gold was jet sprayed onto the glass fiber. DON-BSA and donkey anti-mouse IgG were lined on a nitrocellulose membrane as the test line and the control line, respectively. Sample pad, gold pad, nitrocellulose membrane and absorbent paper were assembled and cut into detecting card. The results showed that the visual detection limits of rapid assay for pure DON standards and DON-spiked sample were 100 μg/kg and 500 μg/kg, respectively. The strips did not cross- react with 21 biotoxins except for 15-acetyl-DON. The strips also exhibited high stability and repeatability. Analysis of DON in wheat and maize samples revealed that data obtained from the strips test were in a good agreement with those obtained from liquid chromatography–tandem mass spectrometry(LC-MS/MS). So it is ideally suited for rapid testing of DON on site.
