摘要
目的探讨超声辐照血卟啉单甲醚和声诺维对乳腺癌细胞株MDA-MB-231增殖及侵袭能力的影响。方法将对数生长期的MDA-MB-231细胞制成细胞悬液,加入HMME和SonoVue后用超声进行辐照。分3组:对照组,不接受治疗;辐照组1:输出声强0.3W/cm2,辐照时间60s;辐照组2:输出声强0.6W/cm2,辐照时间30s。采用MTS法绘制细胞生长曲线,平板克隆方法观察细胞形成克隆的能力,并用Transwell小室法检测细胞侵袭能力。结果对照组的生长曲线陡直,而辐照组的曲线平缓。接种2周后对照组出现肉眼可见的细胞克隆,而辐照组未出现。结晶紫染色后观察,辐照组仅可见由少许细胞聚集成的细胞团。接种后24h、48h观察,对照组、辐照组1、辐照组2穿越小室的细胞数分别为42.00±13.52、3.52±1.26、2.75±0.96(P<0.05)及104.25±12.61、36.25±3.40、28.75±4.99(P<0.05)。对照组穿越小室的细胞数显著高于辐照组(P<0.05)。结论超声辐照联合HMME及SonoVue能有效抑制MB-231细胞增殖,降低细胞形成克隆的能力,并显著降低细胞的侵袭能力。
Objective To investigate the impact of synergy of hematoporphyrin monomethyl ether (HMME) and SonoVue under ultrasound irradiation on proliferation and invasive ability of MDA-MB-231 cell line. Methods Cell suspen- sion of MDA-MB-231 cells in logarithmic growth phase, mixed with HMME and SonoVue underwent ultrasound irradia- tion. There were 3 groups, i.e. control group (without irradiation), irradiation group 1 (sound intensity 0.3 W/cm2 , irra- diation time 60 s) and irradiation group 2 (sound intensity 0.6 W/cm2 , irradiation time 30 s). Growth curve obtained by MTS method was drawn, plate clone formation assay was performed, and changes of invasive ability after irradiation with Transwell assay were detected. Results Irradiation groups showed a flat curve, while control group had a sharp rise curve. Cell clones visible to naked eyes emerged 2 weeks after inoculation in control group, but not in irradiation groups. After crystal violet staining, only cell clusters composed of small amount of cells were seen in irradiation groups. The number of cells transited through matrigel were 42.00+13.52, 3. 524-1.26, 2. 754-0.96 in control group, irradiation group 1 and ir- radiation group 2 24 h after inoculation, and 104. 254-12.61, 36.25+3.40, 28.75+4.99 48 h after inoculation, respec- tively (all P(0.05). The number of cells transited through matrigel of control group were higher than that of irradiation groups (P(0.05). Conclusion Synergy of HMME and SonoVue under ultrasound irradiation could inhibit proliferation of MB-231 cells, reduce cell clone formation and attenuated cell invasive ability effectively and significantly.
