摘要
以甜瓜品种‘新银辉’为试验材料,利用cDNA-AFLP技术分离并获取了甜瓜自毒作用相关水通道蛋白基因片段序列,利用RACE技术获得该基因全长cDNA序列,包含750bp的完整阅读框,编码250个氨基酸,具有6个跨膜区,2个高度保守的水孔蛋白基序NPA以及MIP特征序列,命名为:CmTIP(GenBank登录号:KF134811)。Blastn分析显示该基因与黄瓜CsTIP1;1基因同源性较高;荧光定量PCR分析结果显示该基因在根系分泌物胁迫处理6d时表达量最高,这与cDNA-AFLP分析结果一致。
Differential fragments of tonoplast intrinsic protein (TIP) were analyzed by the cD- NA-AFLP technique and obtained from the leaves of melon, which associated with autotoxicity using 'New Silverlit' as the test material, which was cloned by RACE technology. The full-length cDNA sequence of CmTIP had an ORF of 750 bp and encoded 250 amino acids. Six transmembrane domains, two highly conserved NPA motifs of aquaporins and the characteris- tics sequence of the MIP family were found in CrnTIP. Therefore, the gene of melon could be named as CmTIP(GenBank accession number. KF134811). The results showed that this frag- ment was highly associated with TIP1 ;1 genes of cucumber; the result of fluorescence quanti- tative PCR showed that the gene expressed highest on the 6th day after root exudates treat- ment, which was consistent with the results of cDNA-AFLP.
