摘要
目的观察"标本配穴"电针对高脂饮食诱导的胰岛素抵抗(IR)模型大鼠下丘脑中磷酸化糖原合成酶激酶3β(p-Ser9-GSK3β)表达的影响。方法将30只雄性Wistar大鼠随机分为正常对照组、模型组、"标本配穴"电针组各10只。正常对照组给予普通饲料喂养,另2组给予高脂饲料喂养诱导成胰岛素抵抗模型。"标本配穴"电针组大鼠给予电针刺激关元、足三里、中脘、丰隆穴,每周5次,持续8周。治疗前后检测大鼠体重(BW)、空腹血糖(FBG)、血浆胰岛素(FINS)水平,并计算胰岛素抵抗指数(HOMA-IR);采用免疫组化法检测各组大鼠下丘脑组织中磷酸化GSK-3β(Ser9位点)蛋白的表达。结果高脂饲料喂养8周时,与正常对照组比较,模型组大鼠BW、FBG、FINS、HOMA-IR均显著升高(P<0.01)。经8周电针治疗后,与正常对照组比较,模型组大鼠BW、FBG、FINS、HOMA-IR、p-GSK3β蛋白表达均显著升高(P<0.01);与模型组比较,"标本配穴"电针组大鼠BW、FBG、FINS、HOMA-IR、p-GSK3β蛋白表达均显著下降(P<0.05,P<0.01)。结论 "标本配穴"电针治疗可有效减轻体重及调节葡萄糖代谢紊乱,显著下调胰岛素抵抗大鼠下丘脑磷酸化GSK3β蛋白的表达,这可能是电针改善胰岛素抵抗的机制之一。
Abstracts : Objective To explore the treatment effect of electro - acupuncture with "biao ben pei xue "prescription on the expression of p -Set9 -GSK3β in the hypothalamus of high -fat diet induced insulin resistance(IR) rats. Methods Thirty wistar rats(male) were ran- domly divided into normal control group( NC ), model group (Model)and electro -acupuncture with "biao ben pei xue" prescription group (EA). 10 normal rats were fed with standard diet while the latter two groups were fed with high fat diet to induce IR rats model. The acu- points of Guanyuan ( GV4), Zusanli ( ST36 ), Zhongwan ( CV12 ) and Fenglong (ST40) were used for electro - acupuncture treatment, five times per week for 8 weeks. Detect the level of body Weight( BW), fasting blood glucose( FBG), fasting insulin( FINS), and calculate the value of homeostasis model assessment - insulin resistance ( HOMA - IR) before and after electro - acupuncture treatment. At last, p - Set9 - GSK3β in the hypothalamus was detected by immunohistochemistry. Results Compared with the NC in the 8th week, the BW, FBG,FINS, HOMA - IR in Model were significantly increased (P 〈 O. O1 ). Compared with the NC after electro - acupuncture treatment, the BW, FBG, FINS, HOMA -IR, p -GSK3J3protein expression in Model were significantly increased (P 〈 O. O1 ). Compared with the Model after electro -acupuncture treatment, the BW, FBG, FINS, HOMA -IR, p -GSK3βprotein expression in EA were significantly decreased (P 〈 O. O1 or P 〈 O. 05). Conclusion The" biao ben pei xue" prescription electro - acupuncture treatment can lose the weight, regulate glucose metabolic disturbance, low the p - GSK3βprotein expression in hypothalamus of IR rats, which may be one of the central mechanisms of electro - acupuncture treatment in IR.
