摘要
目的探讨未洗涤红细胞血红蛋白琼脂糖凝胶电泳的可行性。方法以洗涤前后的红细胞分别制备血红蛋白液,同时进行血红蛋白电泳;以血浆代替红细胞加等量溶血剂后在血红蛋白电泳凝胶上电泳。结果 121份静脉血血红蛋白电泳结果:红细胞洗涤前后电泳图目视检查,区带数和位置完全一致,结果间的差异没有统计学意义(P>0.05)。184份脐带血血红蛋白电泳结果:红细胞洗涤前后电泳图目视检查,区带数和位置完全一致,结果间的差异没有统计学意义(P>0.05);血浆代替红细胞电泳结果:13份静脉血浆和13份脐带血浆的电泳图目视检查,在扫描区域内均未见到任何区带。结论未洗涤红细胞可代替洗涤红细胞用于血红蛋白琼脂糖凝胶电泳。
Objective To explore the feasibility of the hemoglobin agarose gel electrophoresis with not washed erythrocyte. Methods 1)Preparing hemoglobin solution with erythrocyte before and after washing, respectively and proceeding hemoglobin elec-trophoresis simultaneously.2)Replacing erythrocyte with plasma and adding equivalent lysing reagent, and then performing elec-trophoresis on hemoglobin electrophoresis gel. Results 1)The hemoglobin electrophoresis results of 121 samples of venous blood:The number and location of the belts on the electrophoregram of the erythrocyte before and after washing are completely consistent with each other by the visual inspection, the differences of the results are not statistically significant (P>0.05). 2) The hemoglobin electrophoresis results of 184 samples of umbilical blood:The number and location of the belts on the electrophoregram of the ery-throcyte before and after washing are completely consistent by the visual inspection, the differences of the results are not statisti-cally significant (P>0.05).3)The electrophoresis results when the plasma instead of erythrocyte:When the electrophoregrams of 13 venous plasmas and 13 umbilical plasmas were visually inspected, there are not any belt can be seen in the scanning area. Con-clusion Non-washed red blood cells can replace the washed red blood cells for hemoglobin agarose gel electrophoresis.
