摘要 目的探讨转化生长因子β1(TGF-β1)基因多态性与妊娠期糖尿病(GDM)发病的相关性。方法选择80例GDM孕妇(GDM组)和80例健康孕妇(对照组),采用DNA直接测序法对TGF—B1基因的+869T/C位点单核苷酸多态性(SNP)进行检测。结果在所有受试者中,TGF—β1基因+869T/C位点存在TT、Tt、CC3种基因型,分布差异符合Hardy—Weinberg平衡,具有群体代表性。GDM组与对照组TGF-β1基因+869T/C位点TT、TC、CC基因型频率及T、C等位基因频率比较差异均有统计学意义(X^2值分别为9.27、9.87,P〈0.05)。结论TGF-β1基因+869T/C位点被证实存在多态性,TGF—β1基因+869T/C位点SNP可能与GDM的遗传易感性相关。 Objective To investigate the correlation between transforming growth factor-J31 (TGF-[31) gene polymorphism and the pathogenesis of gestational diabetes mellitus (GDM). Methods Eighty women with GDM ( GDM group) and 80 normal pregnant women (control group) were studied. Single nucleotide polymorphism (SNP) of TGF-β1+ 869T/C was determined by using direct DNA sequencing. Results In all subjects there were three genotypes including TT, CT and CC. The distribution of these genotypes was found to obey Hardy-Weinberg equilibrium with representation. The differences in the frequency of TT, TC and CC at TGF-β1 + 869T/C were significant, and the frequency of T and C alleles between two groups was also statistically significant (X^2 value was 9.27 and 9.87, respectively, both P 〈 0.05 ). Conclusion TGF-β1 + 869T/C polymorphisms have been confirmed. TGF-β1 + 869T/C SNP may be associated with hereditary susceptibility of GDM.
