摘要
目的研究人源性混合冰冻血小板对wistar大鼠急性创面愈合的作用。方法以手工分离制备的新鲜血小板为原材料,将终浓度2%DMSO冰冻保存3个月的血小板解冻后制备凝胶(Frozen PG);Wistar大鼠15只,脊背两侧做4个直径为1.3 cm的全层皮肤缺损,按照随机的原则分别外敷凝胶纱布[A组(n=15)]、新鲜血小板纱布[B组(n=15)]、重组人表皮生长因子凝胶(rhEG)F纱布[C组(n=15)]、生理盐水纱布[D组(n=15)]于实验的d1、d3、d5、d7、d11更换辅料并随机处死1只大鼠作组织取材,计算创面愈合率及作苏木精-伊红染色(HE)和免疫组织化学染色观察。结果各组创面愈合率均随时间的推移逐渐升高,A、B组创面红润、干净、无渗出物、结痂早、肉芽组织生长丰富,创面触之易出血,愈合后创面平整;伤后d1、d3和d5愈合率分别为A组44.8%,62.8%,69.4%、B组45.0%,58.4%,69.7%、C组39.0%,52.4%,63.0%、D组45%,58.2%,70.2%(P>0.05),d7d时A组为89.4%、B组86.8%、C组83.1%、D组82.8%(P<0.05)。组织学观察显示A、B组在血管生成、炎症细胞的浸润、细胞排列等方面优于C、D组,CD31的表达比C、D 2组高,CD68的表达比C、D 2组出现早,并且表达量高于C、D两组。结论冰冻血小板应用于大鼠创面愈合具有与新鲜血小板同样的促愈合作用。
Objective To investigate the efficacy of frozen platelets prepared from mixed human-derived blood on a- cute wound healing of wistar rats. Methods Frozen platelet gel( Frozen PG) was separated from whole blood and frozen with 2% DMSO for as long as 3 months. A total of 15 wistar rats with four 1.3-cm-diameter circular whole-thickness skin defects in the left and right sides of each rat's back were involved in this study. The defects in each rat were randomly divid- ed into four groups, forzen platelet gel gauze (group A, n = 15 ), fresh platelet gel gauze (group B, n = 15 ), rhEGF gauze (group C,n = 15) and sterile saline gauze (group D,n = 15) . At days 1,3,5,7,11 after operation, dressing were changed and tissue specimen from a sacrificed rat were cutted and inves- tigated by HE and immunohistochemical assay. The size of wounds were measured and the healing rates were calculat- ed. Results The, wounds gradually healed with the passage of time. In group A and B, the wounds were ruddy, clean, no exudation, crusting early, granulation tissue rich, easy bleeding and smooth. There were si^nficant differfences (P 〈 0.05 ) in the wound healing rates among four groups at day 7 but not at day 1,3,5 (P 〉 0.05). Histological exmination showed that the healing effect of group A and B were sigficantly better than group C and D in new vesses generation,inflammatory cell infiltration, cells arrangement, higher expression of CD31, CD68 molecules and earlier expression of CD68 molecule. Conclusion Frozen PG can promote wound healing and shows the e- qual healing effect with Fresh PG.
