摘要
目的探讨膈下逐瘀冲剂对输卵管炎性不孕大鼠NF-κB(核转录因子)、IFN-γ(干扰素-γ)蛋白表达的影响。方法选取120只成熟的雌性大鼠,随机分为6组,正常组20只,其余100只用混合菌注射到大鼠的输卵管中,复制出大鼠慢性输卵管炎性不孕的模型,进行适应性的喂养,分别在第15天、20天后在造模鼠中每组随机选取两只大鼠肉眼及光镜下观察输卵管组织,以判断造模是否成功。然后将造模成功剩余的80只大鼠随机分成模型组、膈下逐瘀冲剂高、中、低剂量组、西药组,每组16只。每组用相应的药物进行灌胃30 d后,处死、取材,用免疫组化法检测输卵管组织中γ-IFN、NF-κB蛋白表达量的变化情况。结果模型组输卵管上皮细胞NF-κB、γ-IFN阳性表达明显高于正常组、膈下逐瘀冲剂高、中、低剂量组、西药组,差异显著,具有统计学意义(P<0.05),膈下逐瘀冲剂高剂量组输卵管上皮细胞NF-κB、γ-IFN阳性表达明显低于中、低剂量组,差异显著,具有统计学意义(P<0.05),而中、低剂量组无明显差异,不具有统计学意义(P>0.05)。结论膈下逐瘀冲剂能够调节输卵管上皮细胞NF-κB、γ-IFN的表达,达到消除炎症的目的,是治疗慢性输卵管炎性不孕有效冲剂。
Objective To explore the impacts of Gexia Zhuyu Infusion on the protein expressions of NF-κB and IFN-γin the rats of oviduct inflammatory infertility.Methods One hundred and twenty ma-tured female rats were collected and randomized into 6 groups.There were 20 rats in the normal group and the rest 1 00 rats were injected with mixed bacteria into fallopian for the duplication of chronic oviduct inflamma-tory infertility models.They were fed adaptively.Separately on the 15th and 20th days,2 rats were randomized from each group from the models for the observation of oviduct tissue with naked eyes and under optic micro-scope so as to judge the success of modeling.The rest 80 rats of successful modeling were randomized into a model group,high-dose group,middle -dose group and low-dose group of Gexia Zhuyu Decoction and western medicine group,1 6 rats in each one.The gastric infusion with corresponding medicine was given in each group.The rats were sacrificed 30 days later for the tissue collection.IHC method was applied to detect the protein expressions of NF-κB and IFN-γin oviduct tissue.Results The positive expressions of NF-κB and IFN-γin oviduct epithelial cells in the model group were higher obviously than those in the model group,high-dose,middle-dose and low-dose decoction groups and western medicine group,indicating the significant difference（P〈0.05）.The expressions in the high-dose group were lower apparently than those in the middle-dose and low-dose groups,indicating the significant difference（P〈0.05）.The difference between the middle-dose and low-dose groups was not significant（P〉0.05 ）.Conclusion Gexia Zhuyu Infusion regulates the expressions of NF-κB and IFN-γin oviduct epithelial cells and removes the inflam-mation.It is the effective infusion in the treatment of chronic oviduct inflammatory infertility.
