摘要
环介导等温扩增(LAMP)是利用设计的4条特殊引物和具有链置换活性的DNA聚合酶,在恒温条件下特异、高效、快速地扩增DNA的新技术。研究选取沙门氏菌编码DNA解旋酶B亚单位的gyrB基因设计了1套引物,总反应体系25μL,63℃恒温1 h,用LAMP对9株沙门氏菌属扩增的结果均为阳性,而大肠杆菌O216等7株致病菌株扩增的结果均为阴性;LAMP的灵敏度为6.8×101cfu/mL。LAMP检测方法更快速、灵敏,有着较为广泛的发展前景。
Loop-mediated isothermal amplification (LAMP) is a new technology which could amplify DNA specifically, efficiently and fast in the constant temperature conditions by four specially designed primers and a chain displacement activity of DNA polymerase. One pair of primer was designed depending on Salmonella gyrB gene sequence which have been reported in genbank. The reaction was run under 63℃ for 1 hour. The amplication result of 9 Salmonella strains were positive and the rest 7 E. coli O216 negative; With sensitivity of 6.8 × 10^1 euf/mL, the detecting way by LAMP is more fast and sensitive and has a brighter development prospect.
