摘要
目的:比较研究白芍、赤芍对环磷酰胺致血虚小鼠的补血作用及机制。方法:采用环磷酰胺法复制小鼠血虚模型,灌胃白芍水提物、赤芍水提物,检测小鼠外周血象、胸腺与脾脏指数及分离血清检验白细胞介素-3(IL-3)、肿瘤坏死因子-α(TNF-α),比较二者的补血作用及机制。结果:白芍高剂量组和赤芍高剂量组均能升高白细胞数(P<0.001,P<0.01),两组之间有明显差异(P<0.05),白芍优于赤芍;白芍高剂量组的胸腺指数有明显升高作用(P<0.001),赤芍高剂量组的胸腺指数无升高作用,二者之间相比,白芍优于赤芍(P<0.001);白芍高剂量组与赤芍高剂量组对IL-3的升高作用均非常明显(P<0.001);白芍中剂量组和赤芍中剂量组对IL-3升高亦有作用(P<0.001,P<0.05),两组之间相比差异显著(P<0.01),白芍优于赤芍;白芍高剂量组降低TNF-α的作用非常明显(P<0.001),而赤芍高剂量组对TNF-α则无明显作用,二者相比差异非常显著(P<0.001),白芍优于赤芍。结论:白芍对环磷酰胺模型的补血作用略优于赤芍,其机制可能为白芍通过对IL-3、TNF-α的共同影响,促进早期造血干细胞的增殖、发育以及调节机体的免疫功能来改善血虚状态;而赤芍的补血机制仅通过影响IL-3促进早期造血干细胞的增殖与发育有关。
Objective: To compare the effects and mechanism of blood enriching on mouse model of blood deficiency syndrome induced by cyclophosphamide of White Peony Root and Red Peony Root. Methods: The mouse model of blood deficiency syndrome induced by cyclophosphamide, White Peony Root and Red Peony Root extract were established. The amount of RBC, WBC, and index of thymus gland and spleen, and the changes of IL-3 and TNF-α in serum were detected after the treatment. Results: Compared with the model group, the amount of WBC in the group of 2g/kg of White Peony Root and 2g/kg of Red Peony Root were increased obviously (P〈0.001, P〈0.01), which had a significant difference compared with the same dose (P〈0.05). The index of thymus gland in the group of 2g/kg White Peony Root was superior to the model group (P〈0.001), the difference was significant compared with the same dose of Red Peony Root (P〈0.001). The IL-3 in serum in both group of 2g/kg of White Peony Root and 2g/kg of Red Peony Root increased obviously (P〈0.001); The IL-3 in serum in both group of lg/kg of White Peony Root and lg/kg of Red Peony Root also increased (P〈0.001, P〈0.05), which showed obvious difference compared with the same dose group (P〈0.01), White Peony Root was superior to Red Peony Root. The content of TNF-α in group of 2g/kg White Peony Root was reduced (P〈0.001), which showed the obvious difference compared with the same dose group (P〈0.001). Conclusion: White Peony Root had a better effect of blood enriching than the Red Peony Root. The probable mechanism was that White Peony Root could not only promote the Hematopoietic stem cell (HSC) proliferation but also regulate the immune function by adjusting the content of IL-3 and TNF-αcommonly. The mechanism of Red Peony Root probably just promoted the HSC proliferation by IL-3.
