摘要
采用构建细菌16S rRNA基因文库研究郎酒超高温大曲中细菌群落结构。随机挑选58个阳性克隆子进行测序分析。结果表明,大曲内细菌群落组成较为简单,主要分布于芽孢杆菌科和高温放线菌科;将序列相似度大于或等于97%的定义为同一个OTU(Operational taxonomic units),可分为6个不同的OTUs。其中,枝芽孢杆菌属(Virgibacillus)和高温放线菌属(Thermoactinomyces)为优势种群,占全部克隆子的比例分别为44.83%和43.1%;其他类群包括芽孢杆菌属(Bacillus)和葡萄球菌属(Staphylococcus)。
The structure of bacterial communities of ultrahigh-temperature Langjiu Daqu was studied through the construction of bacteria 16S rRNA gene clone library. 58 positive clones were randomly selected for sequencing analysis. The results showed that, bacterial biodiversity of Daqu was quite simple and mainly distributed in Bacillaceae and Thermoactinomycetaceae, and 58 positive clones could be divided into 60TUs ( the similarity of different sequences higher than or equal to 97 % is defined as the same OTU).
