摘要
目的:采用反相高效液相色谱建立同时测定积雪草提取物中羟基积雪草苷、积雪草苷、羟基积雪草酸及积雪草酸4种主要成分的分析方法。方法:采用Agilent 1100高效液相色谱仪系统,ZORBAX Eclipse XDB -C8色谱柱(4.6 mm ×150 mm,5μm);流动相为1 mmol/L磷酸二氢钾水溶液-乙腈,梯度洗脱;检测波长205 nm。结果:羟基积雪草苷、积雪草苷、羟基积雪草酸及积雪草酸的色谱响应线性范围良好(R2≥0.9998);平均回收率分别为97.4%、93.7%、97.5%、99.8%,相对标准偏差值分别为3.4%、1.4%、4.7%、4.4%(n=9)。结论:采用反相高效液相色谱建立同时测定羟基积雪草苷、积雪草苷及其苷元的方法高效灵敏、重现性好,可以作为积雪草提取物质量控制的检测方法。
Objective: To establish a HPLC method for simultaneous determination of four major constituents ( madecassoside , asiaticoside , madecassic acid and asiatic acid) in Centella asiatica ( L.) urban extracts .Methods: The analysis was performed on an Agilent 1100 HPLC system with a ZORBAX Eclipse XDB-C8 column (4.6 mm × 150 mm, 5μm) .The four major constituents were separated with gradient mobile phase that consists of 1mmol/L potassium dihydrogen phosphate and acetonitrile at the detection wavelength of 205 nm.Results: The four major constituents all had good linear response in the determination ranges (R2≥0.9998).The average recoveries (n=9) were 97.4%, 93.7%, 97.5%and 99.8%with RSDs of 3.4%, 1.4%, 4.7%and 4.4%, respectively.Conclusion:The developed method is sensitive and has good reproducibility , which can be used as a reference for quality control of Centella asiatica ( L.) urban extracts .
