摘要
目的:探讨芡实对糖尿病肾病大鼠肾组织GLUT1及TGF-β1表达的影响。方法:雄性SD大鼠60只,随机选取10只为正常对照组(N组);其余采用一次性腹腔注射链脲佐菌素(STZ,45 mg/kg)制作DN模型,造模成功后随机分为糖尿病肾病组(DN)、芡实小剂量组(EL,1.5 g·kg-1·d-1)、中剂量组(EM,3.0 g·kg-1·d-1)、大剂量组(EH,6.0 g·kg-1·d-1)及氯沙坦钾组(LP,30 mg·kg-1·d-1),均采用灌胃给药,N组和DN组给予等量蒸馏水。12周后检测大鼠生化指标;HE、Masson染色及电镜观察肾脏病理变化;免疫组化、western-blot法测定GLUT1和TGF-β1在肾组织表达情况。结果:实验12周末,与正常组比较,DN组大鼠肾小球肥大、系膜基质增多,BUN、Scr、24 h尿蛋白定量显著升高(P<0.05),肾组织GLUT1、TGF-β1表达明显上调(P<0.05);与DN组比较,EM、EH组大鼠病理改变减轻,BUN、Scr、24 h尿蛋白定量显著下降(P<0.05),同时肾组织GLUT1、TGF-β1表达下调(P<0.05)。EM、EH组及LP组降尿蛋白作用差异无统计学意义。结论:芡实可能通过下调GLUT1及TGF-β1在肾组织的表达水平,从而延缓糖尿病肾病的进展。
Objective:To investigate the effect of euryale ferox on the expression of GLUT1 and TGFβ1 in renal tissues of diabetic nephropathy rats. Methods :60 SD male rats were randomly divided into 6 groups :the normal control group( group N), dia- betic nephropathic model group( group DN), low -dose Euryale ferox group( EL,1.5g·kg-1^·d-1^), medium -dose Euryale ferox group (EM, 3.0g/kg/d), high-dose Euryale ferox group (EL,1.5g·kg-1^·d-1^) and losartan potassium group (EL,1.5g·kg-1^·d-1^). Rats were treated respectively by intragastric administration for 12 weeks. Meanwhile rats of group N and group DN re- ceived the same volume distilled water by gavage. Blood serum crcatinine, blood urea nitrogen and 24 h urinary protein were measured after the research. The renal pathological changes were observed by histochemistry staining and electron microscope. The expression of GLUT1 and TGF- β1 was detected by immunohistochemistry and western blot. Results: Compared with group N, the glomerular be- came hypertrophic and the mesangial matrix increased in group DN. BUN, Scr, 24 h urinary protein also increased significantly ( P 〈 0.05). Meanwhile the expression of GLUT1 and TGF - β1 in renal tissues increased significantly ( P 〈 0.05). Compared with group DN, the pathology changes were relieved, BUN, Scr, 24 h urinary protein as well as the expression of GLUT1 and TGF - β1 de- creased in group EM and group EH(P 〈 0.05). In addition, there was no significant difference in effect of reducing erinary protein in group EM, EH and LP. Conclusion: Euryale ferox may delay the injury in diabetic nephropathy by down - regulating the expression of GLUT1 and TGF - βl.
