纤维素酶水解连翘苷制备连翘脂素的研究 被引量:7

摘要 目的研究连翘脂素的制备工艺。方法采用纤维素酶水解连翘苷制备连翘脂素,以转化率为指标,通过单因素考察pH值、温度、酶用量和反应时间对转化率的影响;采用MS,1H-NMR,13C-NMR鉴定水解产物。结果酶解反应的最适条件为温度50℃、反应介质pH值为5.2的醋酸-醋酸钠缓冲液,酶与底物比为1∶1,反应时间为48 h,核磁图谱证实产物为连翘脂素。结论纤维素酶水解连翘苷制备连翘脂素反应条件温和,工艺简单可靠,适合工业化生产。 Objective Research on the preparation of( +)-phillygeninin. Methods When using cellulase to facilitate enzymolysis of( +)-phillyrin,the bioconversion rate was taken as the index. By means of single element investigation,elements like pH value,temperature,dosage of enzyme and reaction time are all concerned to see their effects on the conversion rate respectively. MS,1H-NMR,13C-NMR were utilized to identify hydrolysis product. Result The most applicable temperature of enzymatic hydrolysis should be 50 ℃ and reaction medium should be the buffer solution of acetic-sodium acetate with pH 5. 2. The ratio of the amount of enzyme and substrate should be 1∶ 1. The reaction time should be 48h. Analysis of HNMR proved that the reaction product was( +)-phillygeninin. Conclusion During the preparation of( +)-phillygeninin by enzymolysis of phillyrin with cellulose,the reaction was mild and the technology was simple and stable,thus is suitable for industrial production.
