摘要
目的:探讨女贞子是否对MPP+诱导的PC12细胞氧化应激损伤具有保护作用。方法:采用MPP+诱导的PC12细胞作为帕金森病体外模型,同时给予不同浓度的女贞子醇提液、水提液。通过MTT法测定细胞存活率,通过超氧化物歧化酶(SOD)、一氧化氮(NO)、丙二醛(MDA)及乳酸脱氢酶(LDH)试剂盒来检测女贞子对细胞氧化应激损伤的影响。结果:与正常组相比,模型组细胞的细胞存活率显著下降,SOD抑制率、NO含量、MDA含量及LDH水平显著升高(P<0.01);与模型组相比,浓度为1mg/mL的女贞子水提液显著提高细胞存活率,显著降低SOD抑制率、NO含量、MDA含量及LDH水平。结论:女贞子水提液对细胞的氧化应激损伤具有一定的保护作用。
Objective:To evaluate the effect of Fructus Ligustri Lucidi on oxidative stress in vitro. Methods: PC12 cells induced by MPP+ was used as the vitro model of parkinson disease,while adding 70% ethanol extract and aqueous extract of Fructus Ligustri Lucidi of different concentrations. Cell vibility was determined by MTT methods. Effect of Fructus Ligustri Lucidi on oxidative stress was tested by kits of SOD, NO, MDA and LDH. Results:Contrast to normal group, cell viability of model group dropped significant- ly, and inhibition rate of SOD, content of MDA, content of NO and level of LDH of model group significantly raised. Compared with model group, aqueous extract of Fructus Ligustri Lucidi of lmg/mL increased the cell vibility markedly,and lowered inhibition rate of SOD, content of MDA, content of NO and level of LDH significantly. Conclusion : It implys that aqueous extract of Fructus Li- gustri Lucidi can protect PC12 cells from oxidative stress damage.
