摘要
目的:探讨针刺对抑郁症外周和中枢炎性细胞因子的调节作用。方法:采用慢性不可预知的应激方法建立抑郁症大鼠模型。将SD大鼠随机分为正常组、模型组、针刺组和氟西汀组。采用酶联免疫吸附法(ELISA)检测炎性细胞因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的含量。结果:与正常组比较,模型组大鼠血清、海马、额叶皮层IL-1β含量均显著升高(P<0.01),血清、海马、额叶皮层IL-6含量均显著升高(P<0.01),血清、海马、额叶皮层TNF-α含量均显著升高(P<0.01)。针刺治疗可以显著降低模型大鼠血清、海马和额叶皮层IL-1β、IL-6和TNF-α含量(P<0.01)。结论:针刺可以抑制炎性细胞因子,缓解抑郁症外周和中枢炎症反应,这可能是针刺发挥抗抑郁作用的机制之一。
Objective :To observe the effect of acupuncture on inflammatory cytokines response in serum and brain regions of depression. Methods:The chronic unpredictable stress (CUS) rat model with depression was employed in this study. The SD rats were randomly divided into four groups: the normal control, CUS, acupuncture and fluoxetine. The contents of pro - inflammatory cytokines interleukin - 1β ( IL - 1β), IL - 6 and tumor necrosis factor-α(TNF- α) were detected by ELISA. Results:Compared with control group, the contents of IL - 1β in serum, hippocampus and frontal cortex in model group were significantly increased (P 〈 0. 01); the contents of IL- 6 in serum, hippocampus and frontal cortex were significantly increased (P 〈 0. 01 ) ; the contents of TNF - αin serum, hippocampus and frontal cortex were significantly increased (P 〈 0. 01 ). Acupuncture significantly decreased the contents of IL - 1β, IL - 6 and TNF -α in the serum, hippocampus and frontal cortex of the model group (P 〈 0.01 ). Conclusion:Acupuncture treatment could significantly reduce the inflammatory cytokines in serum and brain, and alleviate inflammatory response, which might mediate the antidepression by acupuncture.
