摘要
目的:探讨温阳化瘀方对糖尿病周围神经病变模型大鼠糖代谢紊乱的影响。方法:取SPF级雄性大鼠,随机分为正常对照组、高脂高糖(模型)组、二甲双胍(降糖)组、立普妥(降脂)组、温阳化瘀方18g/kg、12g/kg、6g/kg剂量组。除正常对照组外,其他各组大鼠禁食12h,按60mg/kg剂量尾静脉一次性快速注射链佐菌素(STZ)制备动物模型,并于模型制作当日各给药组大鼠分别ig给予药物治疗,每日一次;同时,均以高脂高糖饲料喂养连续12周。其中降脂组给予立普妥1.25mg/kg,降糖组给予二甲双胍670mg/kg,试药剂量组分别给予温阳化瘀方浓缩液18 g/kg、12g/kg、6 g/kg。正常对照组、模型组给予同样量的生理盐水。每10d称量各组动物体重一次;给药最后一天眼球取血测定空腹血糖;颈动脉采血取血清,用酶联免疫吸附分析方法测定血清瘦素、胰岛素水平,计算胰岛素抵抗指数。结果:温阳化瘀方能降低糖尿病周围神经病变模型大鼠体重,能调节血糖水平,与模型组比较18g/kg、12g/kg、6g/kg剂量组有显著差异。其瘦素含量与模型组比较,均显示有非常显著性差异。能调节胰岛素水平,与模型组比较,均具有非常显著性差异。研究结果表明,本方最小有效剂量为6g/kg,其有效剂量范围是6g^18g/kg)。结论:温阳化瘀方对糖尿病周围神经病变所致体重增加有降低作用,对由此所致血糖、瘦素、胰岛素含量变化能调节到正常水平,提示温阳化瘀方对糖尿病周围神经病变所致糖代谢紊乱有调节糖代谢,治疗糖代谢障碍性疾病的作用。
Objective: To explore the regulatory effects of the Wenyanghuayufang decoction on glycometabolism disorder in diabetic peripheral neuropathy( DPN). Methods: 70 male specified-pathogens free( SPF) Wistar rats were divided into seven groups in randomized manner: the control group with distilled water diet; the model group with high fat( 49% lard) and high sugar( 10% edible sugar); the lipid-lowering group treated with lipitor; the glycemic-lowering group treated with metformin; the 18 g /kg dose,12 g /kg dose and 6 g /kg dose groups treated with the decoction. The rats of every group were taken in high fat and high sugar and taken in addition to the normal control group,a group rats fasting 12h,intravenous single rapid injection preparation of animal model of streptozotocin 60mg /kg by except control group. In addition to the normal control group,other groups of rats were feeding with high fat and high sugar. In addition to the normal control group,other groups and model at all for set of rats given by ig drug treatment,once a day,for 12 weeks. Among them,the rats in lipid- lowering group were taken 1. 25 mg /kg lipitor; the rats in glycemic-lowering group were taken 670 mg /kg metformin; the rats in the decoction large dose group,middle dose group and small dose group were taken 18 g /kg,12 g /kg and 6 g /kg the decoction of concentrated solution separately. The rats in control group and model group were taken normal salt administration by gavage for equal content. The rats’ body weight in every group were weighed; the blood glucose were determinated; the level of serum leptin and insulin were detected by enzyme-linked immunosorbent assay( ELISA). And insulin resistance index( HOMA-IR) was counted. Results: The body weight changed into slow increase; the blood glucose,leptin and insulin content could be regulated to a level of very significant difference( all P < 0. 01) while these groups were compared with the model group after Wenyanghuayu decoction were taken for 12 weeks. This study was demonstrated that the prescription is effective at 6g /kg,and the effective range from 6g ~ 18g /kg. Conclusion: The Wenyanghuayufang decoction can lower body weight and blood glucose,regulate leptin and insulin to a normal level in metabolic syndrome model rats while they were changed with glycometabolism disorder. It was suggested that th decoction be of effects of regulation on saccharide metabolism disorder as diabetic peripheral neuropathy.
