摘要
利用生物信息学和比较基因组学方法对猪、小家鼠、褐家鼠、人、黑猩猩、牛、羊、原鸡、热带爪蟾、非洲爪蟾、斑马鱼、粗尾婴猴、东非狒狒、家猫、狗、野马、普通狨等21个物种的FABP3基因编码区及蛋白质进行分析,探究该基因在种内及种间的遗传分化关系,并全面了解不同物种FABP3蛋白的理化性质、结构、功能位点等。结果表明:21个物种47条编码区共检测到210个多态位点、28种单倍型,不同物种间的核苷酸歧义度、净遗传距离和遗传分化指数的变化范围很大,FABP3基因编码区在种内及种间均存在遗传变异。FABP3蛋白为酸性蛋白,具疏水性,较稳定,是一种定位于细胞质或细胞器(除内质网和线粒体)基质中的非分泌蛋白,其二级结构主要为伊折叠、α-螺旋和自由卷曲。野猪FABP3蛋白的主要结构为10条反平行伊折叠,且有一个重复的+1拓扑结构包绕着一个配基结合位点,属于Calycin结构超家族。其功能保守区为第7~24位氨基酸,推测其主要功能为结合脂肪酸等疏水性分子或有机阴离子。
Aimed to research intraspecific and interspecific evolutionary relationship of FABP3 genes among different species as well as to have a comprehensive understanding of physicochemical property, structure and functional sites of the FABP3 protein, this article analyzed the coding regions and proteins of FABP3 gene by using bioinformatics and compara- tive genomics approach. In our study, 210 polymorphic locis and 28 haplotypes were found and the variation range of the nucleotide divergence, net genetic distance and index of genetic differentiation among 21 species was very apparent. Genetic variation of the coding regions both existed in intraspecific and interspecific. The protein of FABP3 was defined as acidic, non-secreted protein, which was hydrophobic and stable, as well as existed in cytoplasmic or organelle matrix (except for Endoplasmic reticulum and Mitochondria). The secondary structures of the protein mainly contained beta sheet, alpha helix and random coil. It may function to bind Hydrophobic molecules and organic anion.
