摘要
目的:观察诱导型一氧化氮合酶(iNOS)、转化生长因子-β1(TGF-β1)在哮喘大鼠中性粒细胞(PMN)中的表达情况,探讨PMN参与哮喘的可能机制。方法:20只健康雄性SD大鼠随机分成哮喘组和对照组,每组10只,用卵白蛋白(OVA)致敏和激发建立哮喘模型。分离纯化血PMN,免疫组织化学法检测PMN中iNOS、TGF-β1的表达水平。结果:哮喘组血PMN中iNOS、TGF-β1蛋白的表达水平(OD值)分别为0.122±0.017和0.228±0.016,高于对照组(分别为0.076±0.014和0.131±0.015)(均P<0.01)。结论:哮喘大鼠血PMN中iNOS、TGF-β1的表达水平增加,PMN可能部分通过iN0S、TGF-β1合成的增加参与哮喘的气道炎症过程。
Objective: To observe the expressions of inducible nitric oxide synthase (iNOS), transforming groth factor beta 1 (TGF-β1) at blood polymorphonuclear leukocyte (PMN) in OVA-induced rat asthma models and to explore the mechanisms of PMN participating in asthma.Methods: Twenty SD rats were randomly di-vided into a control group and an asthma group, with 10 rats in each group. Rats were sensitized and challenged with ovalbumin (OVA) to establish the asthmatic model. Blood PMN were isolated and puriifed. The expressions of iNOS, TGF-β1 were examined with immunohistochemistry.Results: The levels of iNOS (0.122 ± 0.017, opti-cal density), TGF-β1 (0.228 ± 0.016, optical density) in PMN from the asthma group were signiifcantly higher than from the control group (0.076 ± 0.014, 0.131 ± 0.015) (allP〈0.01).Conclusion: The expressions of iNOS, TGF-β1 at PMN in OVA-induced rat asthma models are signiifcantly increased, so PMN may involved in the pathogenesis of asthma by increasing the levels of iNOS and TGF-β1.
