摘要 目的 探讨髓磷脂碱性蛋白 (MBP)诱导 EAE产生的可能原因。方法 通过酸抽提和 CM-Sephadex50层析 ,分离提取牛 MBP,观察其致 EAE活性 ,用生物活性法检测 MBP刺激的 PBMC上清中 TNF和 IFN的活性。结果 分离纯化得到电泳 1条主带的 MBP,证明其具有致 EAE活性 ,MBP刺激的 PBMC可产生 TNF和IFN。结论 MBP可通过在外周刺激 PBMC产生炎性因子 TNF和 IFN诱导 Objective To explore the possible causes of MBP inducing EAE.Methods By acid abstraction and CM Sephadex50 chromatography purifying MBP and observe the pathogenicity EAE induced by MBP,with bio active assay to test TNF and IFN activity in the supernatant of PBMC stimulation.Results MBP identified by SDS PAGE was a single band .Which showed the activity of pathogenicity of EAE.The PBMC stimulated by MBP could produce TNF and IFN.Conclusions MBP may induce EAE by stimulating PBMC to produce proinflammatory cytokines TNF and IFN in periphery.
