摘要
目的 :研究雷帕霉素 (rapamycin ,RAPA)对小鼠移植排斥反应及免疫功能的影响。方法 :小鼠同种异型耳后心脏移植和皮肤移植 ,二硝基氟苯 (dinitro fluorobenzene ,DNFB)诱导的迟发型超敏反应 (delayedtypehypersensi tivity,DTH)的测定 ,溶血素的测定 ,小鼠腹腔巨噬细胞吞噬中性红实验。 结果 :RAPA可显著延长小鼠同种异型心脏及皮肤移植的存活期 ;显著抑制小鼠DTH反应 ;并使绵羊红细胞 (sheepredbloodcell,SRBC)致敏小鼠溶血素生成显著下降 ;对小鼠腹腔巨噬细胞吞噬中性红作用无明显影响。结论 :雷帕霉素可抑制小鼠同种异型移植排斥反应 ,抑制小鼠细胞及体液免疫功能 。
To investigate the effects of rapamycin on allograft rejection and immune response in mice. Methods:The heterotopic ear heart grafting or skin grafting were done in mice. The delayed type hypersensitivity (DTH) response induced by dinitro fluorobenzene (DNFB), the production of hemolysin of mouse sensitized by sheep red blood cell (SRBC) and the neutral red phagocytosis of the peritoneal macrophage were also tested in mice. Results:RAPA significantly blocked allograft rejection of heart and skin, markedly inhibited DTH response and decreased the production of hemolysin,but had no significant effect on the neutral red phagocytosis of the peritoneal macrophage. Conclusion:RAPA potently blocked allograft rejections in mice and suppressed cellular and humoral immune response, but had no significant effect on phagocytoses of macrophage.
