球囊损伤后血管迁徙平滑肌细胞弹性蛋白酶mRNA的表达 被引量:29

The elastase mRNA expression of migrating medial smooth muscle cells after the damage of balloon angioplasty

摘要 目的 探查血管平滑肌细胞有否弹性蛋白酶mRNA的表达、其在血管组织中的分布及与细胞迁徙的关系。方法 采用球囊损伤大鼠动脉的模型 ,克隆成大鼠胰腺弹性蛋白酶Ⅱ的RNA探针 ,运用原位杂交及免疫组化、电镜等方法。结果 弹性蛋白酶ⅡmRNA的表达见于正常动脉部分平滑肌细胞 ,并因血管损伤而增强。中膜迁徙性平滑肌细胞均有弹性蛋白酶mRNA的强烈表达 ,其细胞周围基质显著减少 ,内弹性板遭到破坏。结论 中膜平滑肌细胞的丝氨酸弹性蛋白酶表达亢进是促进向内膜迁徙并形成平滑肌增生性血管疾病的重要机制之一。 Objective To investigate the expression of the elastase mRNA in vascular smooth muscle, its distribution in vascular system and its relation to cells migrating in morphology. Methods (1) Impaired arteria model: injuring Wistar rats common carotid artery or abdominal aorta with 2F balloon catheter. (2) Cloning the elastase II RNA probe labeled with digoxin( primers which was selected and synthesized from rats elastase II mRNA process RT PCR with rats total pancreas RNA probe. (3) In situ hybridization and immunohistochemistry (paraffin section fixed with 4% paraformaldehyde was employed in hybridization in situ. BrdU immunohistoch emistry dye was processed on the specimen of paraffin section after hybridization in situ). (4) Observe with transmission electron microscopy (Specimen was fixed and perfused with 2.5% glutaralhyed, then fixed with 1% Osmium and 1%tanaic acid, dyed with uranium acetate). Results (1) The elastase mRNA expressed in endothelial cells, adventitia fibroblast and some media smooth muscle cells of the carotid artery and aorta in intact rats. (2) These expression became intensive in the case of artery after balloon injury. (3) The expression of proliferative cells and migrating cells was all intensive. (4) Elastic fibers and collagen fibers in the media were reduced resulting in decreased contact flaques of mSMC with elastic fibers. Conclusion This study suggested that elastase synthesized by mSMCs played an important role in eointima formation after balloon injury.
