摘要
目的:构建口服5氨基水杨酸结肠定位释药系统。方法:以延时性和pH值依赖性为结肠控释依据,采用多层薄膜包衣法制备结肠定位释放片剂;采用 γ射线显影法,考察包衣片在狗体内的释药部位与释药时间。结果:研制的口服5氨基水杨酸结肠定位释药片剂制备工艺简单,药物释放稳定,在狗的升结 肠崩解释药。结论:构建的5氨基水杨酸的口服结肠定位释药系统具有进一步开发应用前景。
:OBJECTIVE To prepare 5 ASA colon drug delivery system (CDDS). METHODS The tablets were coated with a three layered film cons isting of an acid soluble layer,a hydrophilic layer and an enteric layer.The dr ug was prevented from releasing in the small bowel and the onset time of drug re lease in the intestine was controlled by the thickness of the acid soluble laye r , which would dissolve in the inflammatory colon because the local pH decreased to (4.7±0.72).The vivo drug releasing site and time in dogs were inves tigated by γ ray scintigraphy with Tc99 as indicator. RESULTS The technology to make the 5 ASA CDDS is simple and the drug was released stea d ily.The coated tablets were disintegrated in dog's ascending or transverse colon . CONCLUSIONS The 5 ASA CDDS is promising to be further studied and be utilized in the near f uture.
