摘要
目的 :研究苦参碱抑制 Hep G2细胞增殖的机制。方法 :(0~ 1.5 ) g/ L苦参碱作用 Hep G2细胞 3天 ,免疫细胞化学和放免法检测 AFP、PCNA、c AMP、c GMP量的改变 ;亲和层析法分离肝癌 GGT(HSGGT) ,酶动力法检测 HSGGT和 GGT酶活性。结果 :反应细胞分化的指标 c AMP升高 ,c AMP/ c GMP比值升高。反应细胞增殖的指标 AFP、PCNA、c GMP、GGT、HSGGT量均降低。结论 :苦参碱在代谢水平上诱导了 Hep G2细胞分化 ,是苦参碱抑制 Hep G2细胞增殖的机制之一。
Objective:To study the proliferation inhibiting function of matrine induced HepG2 cells.Methods:Matrine(0~1.5 g/L) treated HepG2 for three days,the quantitative changes of AFP,PCNA,cAMP and cGMP were detected with immunocytochemistry and radioimmunoassay;hepatocellulars'GGT(HSGGT)was isolated with affinity chromatography;enzymatic activation of HSGGT and GGT were detected with enzymatic assay.Results:The index of cells differentiation_cAMP rose and the ratio of cAMP to cGMP rose;the index of cells proliferation_AFP,PCNA,cGMP,GGT,HSGGT fell down.Conclusion:Matrine has induced HepG2 cells differentiation at metabolism level.It has the function of matrine-inhibiting proliferation of HepG2 cell.
